Yes. they come out. All seats in every car/truck ever made come out.
They aren't welded to the floor they are bolted in place.
Now, it can be a huge pain to take them out. You might have to peel back the Carpet and unbolt them. In fact I'm almost positive that's the case with the Highlander.
Doesn't it come with Fold Flat seats? So that you can fold them down and the whole Back area will be level?
The seats that can be easily taken out (like in Vans) are called "Quick Release" seats. Nobody makes them for the Highlander.
