It's similar question to "do you want a Corolla or Camry?" Difference is size.
Rav4 is pretty cute, small and maneuverable. My bro's wife has driven one for years and loves it, hates the new design tho' for 2010. But still prefers Rav's.
But brother drives a Highlander, mostly because he's a landscaper, uses it for work, carrying lots of plants and tools and stuff.
The Highlander is a workhorse, But you won't get really good mpg. The Rav is cute and easy going, so you'll get good mpg, but won't be able to carry a whole lot as much as the Highlander, but you can still carry a big bunch of stuff.
Toyota pretty much started the new line of mini-SuV with the Rav4. Everyone else worked to catch up with the Rav. The highlander is more of catching up to Jeep or such. But both are very good toyotas.
You mght try to drive each one, because the will handle differently and something about them you'll find you like, some things you'll hate. It really can matter by driving each one. You might like how the Rav turns easier, but prefer the stability and size of the Highlander.
