Both are very well built, my brother has owned a Tundra since early '08 when he purchased it new, and already has nearly 50k miles on it, including farm use, HEAVY towing (26' Big Game fishing boat on weekends, livestock trailers during the week) and has had no problems.
My uncle owns a 2009 Mustang, which has little mechanical difference from the 2010 model, and has had no issues.
Which is better will depend entirely upon what you need it to do. If you need towing ability and an open cargo space, the Tundra will obviously be better for you. If cargo capacity isn't important, the Mustang will ride better and get a few more mpg over the Tundra.
