First off - the Camry comes with an inline 4, not a V-4. The only car with something close to a V-4 is the Subaru boxer engine - which is really called an H-4 (as in horizontally opposed).
The 2010 V-6 Camry engine offers a significant increase in horsepower, with a pretty small hit to MPG. Unless you really put a premium on MPG (drive lots of miles) I think the trade-off of more power for a few MPG is worth it.
One tidbit I have learned as a long time Camry V-6 owner (1999) - is they are more expensive to maintain. Tune-ups cost more, struts are beefer/cost more, replacing fluids costs more because it holds more, bigger battery - many things cost more, etc. etc. etc. Not huge - but worth considering if you plan to keep the car for awhile.
Here is my recommendation - if you drive mostly in city you won't notice the difference in power, so the 4 is fine.
If you drive mostly highway miles - including Interstate merging, or mountain areas.....get the V-6.
If you drive a TON of miles - MPG and low maintenance cost are important, so getting the 4 banger is probably the best bet.
