They are both excellent vehicles. The Land Cruiser has some extra off-road technology such as crawl control but I would not be able to justify the extra cost.
For family transportation, I would choose the Sequoia. It is based on the excellent Tundra truck and is a versatile, reliable platform.
The only advantage I see of the Land Cruiser is hard-core off-road performance. It has a solid rear axle that has more wheel travel than the independent rear suspension of the Sequoia to help keep the tires on the ground in uneven terrain and some adaptive suspension features.
For general use, I would prefer the Sequoia. It is an excellent vehicle and the cost of purchase, insurance is substantially lower than the current-generation, 200-series Land Cruiser. Maintenance cost is also likely lower with the Sequoia. The 200-series has many, complex, electronic systems that would be very expensive to repair in the event of failure.
