*if* it is an engine knock, the ones we fear, crank, rod, wrist pin, when they first show up, if something hasn't just let go, the engine *typically* runs w/o missing or stumbling. that's *typically*. not always the case.
if it is the feared knock, your description, *comes and goes while shifting*, smacks of a load bearing component. rod main or wrist pin. associated symptom w/this issue is slow to build oil pressure after starting. that is the oil light is slow to go out after start.
however, many other things can sound like a "knock" to the untrained ear. take it in to a reputable shop. any mechanic worth their salt can tell a rod, crank, even wrist pin and valve train issues just standing next to it.
there are minor things like pcv valve clogged and rocker noises that can easily cause stumbling and to the untrained ear, sound like a knock.

on a used vehicle, inspection of oil and coolant levels, and fluid leaks under the vehicle, are most critical, and should be done b4 driving every time till one gains confidence in the vehicle. slacking up these inspections till the point that recommended service intervals are met.
